Data science concepts and considerations in conducting and interpreting exposome-wide association studies.
Abstract
Central to the exposome concept includes data-driven and comprehensive approaches to identify individuals and mixtures of correlates with disease and health outcomes. Here, we attempt to offer a guide for new researchers on how to interpret and evaluate data-driven exposomic studies. We first introduce the concepts of contributions from genes and environment to phenotypes and data-driven analysis in large observation cohorts. We then give an overview of exposome-wide association studies (ExWAS), a versatile analytical approach to identify robust signals from large and complex exposomic data, and discuss their key considerations and extensions. We also briefly cover emerging topics for exposmic studies, including FAIR Data Principles, administrative healthcare datasets, and functional exposomic data. Finally, for any data-driven study associating multiple exposures with a phenotype and involving correction for multiple comparisons, regardless of data types and research questions, we propose to standardize the terminology for this approach as "exposome-wide association study, ExWAS”. This will facilitate effective communication among researchers and improve literature retrieval.
